What is an aunt/uncle (avuncular) DNA test?

An avuncular DNA test determines whether a biological relationship exists between an alleged aunt or uncle and a niece or nephew. The test works because biological siblings share genetic material inherited from the same parents. By comparing the DNA of an aunt or uncle with the child's DNA, the lab can calculate the probability that they are biologically related through a shared parent.

When would I use an aunt/uncle DNA test instead of a paternity test?

An aunt/uncle test is most useful when the alleged father (or mother) is unavailable for direct testing due to death, incarceration, military deployment, refusal, or unknown whereabouts. No, but including her sample strengthens the analysis. Alternative samples go through forensic-level extraction and may take slightly longer. You can combine a cheek swab from one participant with an alternative sample from another in the same test.
