What a discreet aunt or uncle DNA test does
An aunt or uncle DNA test compares genetic markers between an alleged aunt or uncle and a niece or nephew to determine whether the two share a biological relationship through the same paternal or maternal line. Because full siblings share about half their DNA, an aunt or uncle inherits a predictable share of the same markers as the biological parent, which is what makes this comparison scientifically meaningful when the parent is not available for testing.
This kit is the alternative-sample version of that test. One participant provides a standard cheek swab, and the other provides an alternative DNA sample from a used personal item. That flexibility is what makes it work in situations where a cheek swab cannot be collected from one of the two participants.
When families choose this test instead of a direct paternity test
A paternity test is always the more conclusive option when the alleged father is available. The aunt or uncle test is a second-best option, chosen when direct testing is not possible. The most common reasons families choose this route:
-
The alleged biological father has passed away and cannot be tested directly.
-
The alleged father is unreachable, has left the country, or refuses to participate.
-
The child's biological mother is deceased or unwilling to test, and the child needs to work through the paternal side of the family.
-
A close family relationship needs to be verified before pursuing a formal legal or custody process.
-
An adult adoptee wants to confirm a suspected biological tie with an aunt or uncle before contacting other family members.
How accurate is an aunt or uncle DNA test using an alternative sample?
When viable DNA is recovered from both samples, results indicate a strong, moderate, or weak likelihood of the tested relationship. An aunt or uncle test is inherently less conclusive than a direct paternity test because the shared DNA percentage between an aunt or uncle and a niece or nephew is roughly half of what a parent and child share.
Including the biological mother's cheek swab in the case, when possible, meaningfully strengthens results by letting the lab separate maternal from paternal DNA in the child's profile. My Forever DNA guarantees a specific 46 DNA genetic markers on every avuncular test.
The lab uses 46 DNA genetic markers plus dual processing through a US-based AABB-accredited and CAP-certified laboratory partner. If the result comes back inconclusive, the lab can advise on whether additional relatives (grandparents or a second sibling) could be tested to strengthen the answer.
Alternative DNA samples we accept for the discreet participant
The participant who cannot provide a cheek swab can submit any of the following. Sample age, storage, and handling all affect whether viable DNA can be recovered, so following the instructions in the kit matters.
-
Used toothbrush (about 20 or more uses recommended)
-
25 to 50 strands of hair with the root or follicle attached
-
Nail clippings, 5 to 10 pieces
-
Earwax on a cotton swab
-
Chewed sugar-free gum
-
Dental floss picks
-
Used tissue with mucus
-
Razor debris from an electric or disposable razor
-
Cigarette filters
-
Dried blood on a bandage or cloth
Water bottles, cups, utensils, and breast milk are not accepted. They rarely contain enough usable DNA and often carry contamination from other people. If you have an item that isn't on the list, contact us before shipping so we can advise on viability.
When to choose a legal DNA test instead
This kit is for informational use only. The results are not admissible in court and cannot be used for custody, immigration, child support, or estate proceedings. If any legal process is involved or may become involved, a legal chain-of-custody test is the right choice.
Legal testing requires witnessed sample collection, verified photo identification, and a documented chain of custody, which cannot happen from home. We offer legal testing separately at our Omaha, Nebraska office.
