What a grandparent DNA test does
A grandparent DNA test compares genetic markers between one or two grandparents and a grandchild to determine whether they share a biological relationship. Because a child inherits about 25% of their DNA from each grandparent, the test looks for the specific markers a child would carry if the tested grandparent is a biological grandparent through the alleged parent's side of the family.

One grandparent vs. two grandparents: how much stronger is the two-grandparent test?
When both grandparents on the alleged father's side can be tested (the paternal grandmother and grandfather), results are meaningfully stronger than testing a single grandparent. That's because the lab can distinguish which grandparent contributed which markers, which narrows the range of possible results into a clearer likelihood.
A single grandparent test still works and produces a valid likelihood indication, but the result is more likely to come back as "moderate" or "inconclusive" rather than "strong" or "excluded." If both grandparents are available, test both. If only one is available, the test is still worth doing, but expect a wider range of possible outcomes.
When a grandparent test is the right choice vs. an aunt or uncle test
Both grandparent testing and aunt or uncle testing work through the extended family when a direct paternity test isn't possible. Choose the grandparent test when the alleged father's parents are available and willing. Choose the aunt or uncle test when the grandparents aren't available but a sibling of the alleged father is. If none of these relatives are available, the paternity question typically cannot be answered scientifically.

The mother's DNA lets the lab identify which markers in the child came from the maternal side, isolating the paternal-side markers for comparison against the grandparents' DNA.
