    An aunt or uncle DNA test compares genetic markers between an alleged aunt or uncle and a possible niece or nephew to determine whether they share a biological relationship through the same paternal or maternal line. Because full siblings share about 50% of their DNA, an aunt or uncle inherits a predictable share of the same markers as the biological parent. That predictability is what makes the test scientifically meaningful when a direct paternity or maternity test isn't an option.

    The aunt or uncle test is a second-best option compared to a direct paternity or maternity test. Order it when:

    When to add the mother's cheek swab

    Including the biological mother's cheek swab meaningfully strengthens avuncular results by letting the lab separate maternal from paternal DNA in the child's profile. The mother's sample is optional but recommended when possible. Coordinate with your dedicated DNA specialist before mailing if you want to add her.

    Paternity testing is a direct parent-child comparison, which is a relatively simple genetic question. Aunt or uncle testing requires comparing patterns of shared markers across an inheritance model and calculating a likelihood ratio for the relationship. The extra statistical work happens within the 2 to 3 business day window.

    What can extend the timeline

    The most common cause of delay is a swab that arrived at the lab with insufficient DNA. Your specialist contacts you before running the comparison if a swab is low-DNA, and you decide whether to submit fresh samples. Weekends and holidays are not counted in the turnaround window.

    How It Works

    Step 1 · Order and receive the kit

    Kit ships within 1 business day in plain FedEx packaging. US delivery in 1 to 2 business days.

    Step 2 · Collect cheek swabs

    Both participants collect two cheek swabs each by rubbing each swab firmly against the inside of the cheek for 60 seconds. Painless at any age.

    Step 3 · Air-dry the swabs

    Let all swabs air-dry at room temperature for at least 30 minutes before packaging. Never seal wet swabs, never use plastic bags.

    Step 4 · Package samples separately

    Place each participant's dry swabs into their labeled envelope. Complete the notification form with participant identifiers and your contact info.

    Step 5 · Ship samples to the lab

    Put everything into the prepaid FedEx return envelope. Drop at any FedEx location or arrange a pickup. Tracking included.

    Step 6 · Lab logs samples and confirms intake

    Your specialist confirms intake by email or phone.

    Step 7 · Avuncular comparison

    The lab runs the avuncular comparison with dual processing over 2 to 3 business days.
