What a grandparent DNA test does
A grandparent DNA test compares genetic markers between one or two grandparents and a grandchild to determine whether they share a biological relationship. A child inherits approximately 25% of their DNA from each grandparent. The test looks at that inheritance pattern to indicate whether the tested grandparents are the child's biological grandparents through the alleged parent's side of the family.
One grandparent vs. two grandparents
When both grandparents on the alleged parent's side can be tested (paternal grandmother and paternal grandfather), results are meaningfully stronger than testing a single grandparent. Two grandparents let the lab distinguish which grandparent contributed which markers, narrowing the range of possible outcomes into a clearer likelihood. A single-grandparent test still works but the result is more likely to come back as moderate or inconclusive.
If only one grandparent is available, the test is still worth doing. If both are available, testing both is worth the small extra fee for the second grandparent's marker analysis.
When to add the mother's cheek swab
Including the biological mother's cheek swab strengthens the result significantly by letting the lab identify which side of the family the shared DNA came from. The mother's sample is optional but recommended when available. Coordinate with your specialist before shipping if you want to add her.
When to choose this test vs. an aunt or uncle test
Both grandparent and aunt or uncle testing work through the extended family when a direct paternity test isn't possible. Choose grandparent testing when the alleged father's parents are available and willing. Choose aunt or uncle testing when the grandparents aren't available but a sibling of the alleged father is. Grandparent testing is usually the stronger option when it's available, because grandparent-grandchild inheritance is more predictable than avuncular inheritance.
